FWIW, the vaseline will help the o-ring seat itself a little better. If the block (or whatever) is a little sticky to the rubber and the barb as well then it will stretch and twist the o-ring slightly, as in VERY slightly, as it is tightened but it could be enough to cause a slow drip. I agree it shouldnt be necessary but it doesn't hurt.

I would not mix vaseline with o-rings since the petroleum jelly will eat rubber alive.

Yes petroleum based Vaseline is not good with rubber although something like KY jelly is fine.

If you find teflon tape is necessary, be very careful with it. If it gets into your pump, or worse yet the rad, you'll have big trouble.
