would be worth it to go from my Phenom II X3 720 to one of these new six cores when they come out, or should I wait? I mean my 720 runs plenty fast, but 6 cores just sounds cool!

if you need the multithreaded processing power, yes. if you game probably not(most games anyway).

Aren't the Thuban processors supposed to disable cores to make more room to clock up the cores being used? A la turboboost? Should improve single-threaded performance, but we'll wait for a review before knowing too much.
